Background
TFEB protein, which stands for Transcription Factor EB, is a crucial regulator of lysosomal biogenesis and autophagy. It is a member of the MiT family of transcription factors. When TFEB is activated, it translocates to the nucleus and binds to the promoters of various genes related to lysosomal and autophagy functions, thereby promoting the expression of these genes. This process helps cells to enhance their ability to degrade and recycle cellular components, which is essential for maintaining cellular homeostasis, especially under conditions of cellular stress such as nutrient deprivation or the presence of damaged organelles. Abnormal regulation of TFEB has been implicated in several diseases, including neurodegenerative diseases and lysosomal storage disorders, making it a potential therapeutic target.
